Pathogenesis of celiac disease: identification of isopeptides between tissue transglutaminase and gluten peptides from wheat, rye and barley by LC-MS/MS

    Abstract


    Human intestinal tissue transglutaminase (TG2) forms covalently linked complexes with gluten peptides by transamidation. Highly purified gluten protein types were isolated and their proteins were characterized and relatively quantified. Model systems and a strategy for the identification of isopeptides were developed and isopeptides between TG2 and model peptides as well as biological gluten peptides of wheat, rye and barley were detected by discovery-driven LC-MS/MS and additionally verified by targeted LC-MS/MS analyses.
