Beschreibung
Unlike standard solid phase synthesis, photolithographic synthesis of DNA uses only moderately acidic reagents, and therefore it is intrinsically less susceptible to fragmentation due to depurination. Nevertheless, synthesis of long oligonucleotides via photolithography has demonstrated lower average yield. Our findings demonstrate chain cleavage of DNA sequences due to guanine depurination and we present methods to optimize synthesis conditions for suppressing depurination thereby resulting in enhanced sequence fidelity and synthesis yield.| Zeitraum | 3 Sept. 2024 → 6 Sept. 2024 |
